摘要
利用微生物Absidiasp.R9g产的芦丁-α-鼠李糖苷酶(RhaR),水解芦丁制备鼠李糖。24.0g芦丁经酶解后,采用大孔吸附树脂对产物进行分离,得到含有4.18g鼠李糖的粗糖液,其得率为17.4%。结合结晶法对鼠李糖粗糖液进行精制,当鼠李糖水溶液质量浓度达到0.4g/mL时,鼠李糖可达到过饱和状态而开始结晶,所得晶体颗粒饱满,无色透明,结晶得率为27.5%。通过高效液相色谱检测,纯度达到99.0%。
The rutin was hydrolyzed into rhamnose by rutin-a-rhamnosidase (RhaR) from Absidia sp. R9g. The hydrolytic product (24.0 g) was separated by macroporous resin and the crude sugar containing 4.18 g rhamnose was obtained with a yield of 17.4%. The rhamnose was further purified by crystallization. The rhamnose begun to crystallize when the concentration was 0.4 g/mL above. The colorless and transparent crystal of rhamnose was obtained with a yield of 27.5 %, and the purity was about 99.0%.
