摘要
目的探讨进食不同浓度锌对脊髓损伤(spinal cord injury,SCI)大鼠BDNF表达的影响。方法取60只雄性SD大鼠,建立Allen's SCI模型,随机分成假手术组、模型组、低锌喂养组和高锌喂养组。HE染色检测各组大鼠脊髓损伤情况,实时定量PCR检测各组大鼠脊髓中BDNF表达的变化,BBB评分检测后肢运动功能的改善情况。结果高锌喂养组BDNF表达量在每个时间点均明显高于其他3组,组间差异有显著性意义(P<0.01)。术后1 w脊髓HE染色显示高锌喂养组脊髓损伤减轻;术后各时间点,高锌喂养组大鼠BBB评分明显高于模型组和低锌喂养组。结论进食合适浓度锌可促进SCI大鼠BDNF的高表达,并明显改善大鼠后肢运动功能。
Objective To investigate the influence of supplement zinc on expression of BDNF in rats with spinal cord injury. Methods Allen's injury model of T10 spinal cord was successfully established in SD rats. 60 rats were randomly and evenly divided into four groups. Rats in the zinc - deficient dietary group ( ZD), model group and zinc - high dietary group (ZH) were given free access to purified diet respectively. Rats that had sham operation were treated with laminecto- my at the T10 vertebrae, but their spinal cord was not stricken and they were fed by 30 mg Zn/kg. Neurological function was scored by BBB test. The spinal cord segments were gathered for histologic examination. Real - time PCR were used to detect the expression of BDNF in each group after SCI. Results In ZH group, the expression of BDNF was significantly increased compared with that of other groups (P 〈 0. 01 ). The rats in the ZH group had high BBB scores, slight histologic changes compared with those in the ZD and model groups at the three observation time points (P 〈 0. 01 ).
