高脂诱导的非酒精性脂肪性肝病伴胰岛素抵抗大鼠模型建立方法 被引量:1

摘要 目的建立一种简单易行且符合人类疾病特征的大鼠非酒精性脂肪性肝病伴胰岛素抵抗动物模型。方法将18只SD大鼠随机分为两组,正常对照组大鼠给予基础饲料喂养,模型组给予高脂饲料及18%蔗糖水喂养;观察并记录两组大鼠的一般情况及体重变化,12周后取材检测大鼠的血清血脂、血糖、胰岛素、转氨酶,检测肝脂、丙二醛、肝脏HE染色病理学指标。结果模型组大鼠的血脂、肝指数、肝脂、胰岛素抵抗指数明显高于正常组,病理学检查提示模型组大鼠肝组织呈脂肪变和炎性变。结论科学比例的高脂配方可以成功诱导建立非酒精性脂肪性肝病伴胰岛素抵抗大鼠模型。 Objective To establish a simple and in accordance with human disease characterized nonalcoholic fatty liver disease animal model of insulin resistance. Methods 18 SD rats were randomly divided into two groups,normal control group rats were given basic chow,model group were given high fat diet and 18% sucrose sucrose feeding;the rats general and weight changes in two groups were observed and recorded,the drawn serum lipids,glucose,insulin,ALT, liver fat detection,MDA,HE staining of liver pathology indicators was detected respectively after 12 weeks. Results The lipids,liver index,liver fat,insulin resistance index in model group was significantly higher than that of the normal group.Pathology revealed,model group in the rat liver tissue was fat and inflammatory change. Conclusion Science proportion of high-fat recipes can be successfully induced to establish nonalcoholic fatty liver disease associated with insulin resistance in rats.
