摘要
目的探究磷钨杂多酸与牛血清白蛋白(BSA)的相互作用机制。方法利用UV-VIS吸收光谱与荧光光谱,研究磷钨杂多酸与BSA的猝灭类型与作用机制,结合同步荧光法考察磷钨杂多酸对BSA构象的影响。结果磷钨杂多酸对BSA的荧光猝灭为静态猝灭,其结合常数分别为6.54×103(298K)与3.67×103(310K),结合位点数n为1,与BSA相互作用方式为静电引力。同步荧光法测定结果显示,磷钨杂多酸与BSA发生了相互作用,但没有改变BSA的构象。结论本研究通过分析磷钨杂多酸与BSA之间的作用机制,为后续多酸药物的研发提供了理论依据。
Objective To study the interaction of phosphotungstic heteropoly acid and bovine serum albumin (BSA). Methods The quenching type and mechanism of action between phosphotungstic heteropoly acid and BSA has been studied by UV-VIS absorption spectrum and fluorescence spectrum,the conformation of BSA impacted by phospho- tungstic heteropoly acid was also discussed by synchronous fluorescence. Results The fluorescence quenching of phos- photungstic heteropoly acid with BSA was static quenching and the binding constants at 298K and 310K were 6.54x 10~ (298K) and 3.67x103 (310K) respectively.The number of binding site (n) was 1.The interaction of phosphotungstic heteropoly acid with BSA was electrostatic force.The result of synchronous fluorescence revealed that phosphotungstic heteropoly acid interacted with BSA without changing its conformation. Conclusion The mechanism of action of phos- photungstic heteropoly acid and BSA was studied in this test which provided a theoretical basis for further research on heteropoly acid drugs.
