胆囊收缩素-8、β-内啡肽与多巴胺在大鼠癫痫发作中的相互关系

摘要 采用核团微量注射、放射免疫分析及荧光分光测定等实验方法 ,探讨了大鼠癫痫发作过程中胆囊收缩素 8、β 内啡肽和多巴胺的相互关系及作用。结果表明 :(1 )海马内注射胆囊收缩素 8,大鼠癫痫发作明显减轻 ;海马内注射L 365和胆囊收缩素 8,胆囊收缩素 8压抑癫痫发作的作用消失 ;(2 )癫痫发作后海马内 β 内啡肽含量显著降低而胆囊收缩素 8含量显著增加 ;(3)大鼠海马内注射胆囊收缩素 8,海马内β 内啡肽含量明显减少 ;若大鼠海马内注射胆囊收缩素 8再诱发癫痫 ,此时大鼠癫痫发作明显减轻 ,海马内 β 内啡肽含量显著降低 ;(4)海马注射 β 内啡肽抗血清后大鼠癫痫发作明显减弱 ,海马内胆囊收缩素 8含量明显升高 ;(5)大鼠海马内注射胆囊收缩素 8,海马内多巴胺含量明显增加。以上结果提示 :胆囊收缩素 8具有压抑癫痫发作作用 ,该作用可能是通过胆囊收缩素 B受体而实现的 ;而内源性β 内啡肽可能具有加强癫痫发作作用 ;胆囊收缩素 8与 β Effect of CCK8, βEP and DA on seizure and their relationship were studied by radioimmunoassay and microinjection in rats. The results were as follows: (1)Seizure was significantly decreased by injecting CCK8 in hippocampus CA 3 area (P<01).The effect of CCK8 inhibiting seizure disappeared by injecting L365+CCk8 in hippocampus CA 3 area (P<001); (2)The contents of βEP in hippocampus was significantly decreased (P<005),but that of CCK8 was significantly increased (P<005); (3)The contents of βEP in hippocampus was significantly decreased by injecting CCK8 in hippocampus CA 3 area.Seizure and the contents of βEP in hippocampus were significantly decreased by injecting CCK8 in hippocampus CA 3 area and then inducing seizure(P< 001); (4)Seizure was significantly decreased and the contents of CCK8 in hippocampus was significantly increased by injecting βEP antiserum into hippocampus CA 3 area (P<005); (5)The contents of DA in hippocampus was significantly increased by injecting CCK8 into hippocampus CA 3 area (P<005).The results indicated that CCK8,βEP and DA may have inhibitive relationship and the endogenous βEP may increase seizure.
