用低温诱导法制备人高分辨染色体

Human high resolution chromosome preparation induced by low temperature

目的 :探讨非药物作用获得人高分辨染色体。方法 :外周血淋巴细胞在 37℃中培养 66h ,入 4℃冰箱4h ,再分别入 37℃恢复培养 0、1 0、2 0、30、4 0、50、60min ,终止培养前以低浓度 ( 0 0 8μg/ml)秋水仙素处理培养物 30min。结果 :不恢复培养 ( 0min)或短时恢复培养 ( 1 0、2 0min)组分裂指数最高 ,晚前期、前中期和早中期染色体数目最多。结论 :该法简便 ,不需药物诱导 ,便于推广。

Objective To study the method of obtaining human high resolution chromosome without any medicine.Methods:Peripheral biood Lymphocytes were cultiveted for 66h at 37℃,4h at 4℃ and resumed the culture at 37℃ for 0 min,10min,20min,30min,40min,50min and 60min,respectively.The cells were treated with colchine(0.08μg/ml)for 30min before ended the culture.Results:There were the highest mototic index and the most number of late prophase,prometaphase and early metaphase chromosome among the groups which were resumed for 0 min ,10min and 20min,respectively.Conclusion:The characteristics of this method is simple,convenient,economical and easy to popularize.
