三氧化二砷对恶性转化细胞生长的影响 被引量:2

摘要 目的探讨三氧化二砷对反式苯并芘(anti-BPDE)恶性转化细胞生长的影响,揭示小分子核酸(miRNA)在三氧化二砷抑制anti-BPDE恶性转化细胞生长过程中的作用。方法用不同浓度的三氧化二砷处理恶性转化细胞16HBE-T;利用CCK8试剂及软琼脂克隆形成实验观察三氧化二砷对16HBE-T细胞增殖和克隆生长能力的影响;以miRNA基因芯片检测三氧化二砷处理后细胞中miRNA的表达差异。运用生物信息学工具分析差异表达miRNA的整体功能。结果 25μmol/L三氧化二砷明显抑制16HBE-T的细胞增殖(P<0.05);25μmol/L三氧化二砷明显降低16HBE-T细胞克隆形成(P<0.01)。三氧化二砷处理后,53个miRNA的表达发生改变:11个表达下调,42个表达上调。生物信息学分析这些差异表达miRNA可影响细胞周期和肿瘤生长相关的信号通路。结论三氧化二砷可抑制anti-BPDE恶性转化细胞生长,且这种作用可能与miRNA表达谱的改变有关。
