包裹阿霉素的叶酸-聚乙二醇-聚谷氨酸胶束对肝癌的靶向治疗能力 被引量:4

摘要 目的验证包含抗癌药物阿霉素(DOX)的叶酸-聚乙二醇-聚谷氨酸胶束(folate-PEG-PLGA)的肝癌靶向治疗能力。方法制备含DOX的folate-PEG-PLGA靶向胶束,并检测其DOX含量。采用荧光实验和流式细胞仪对比各组(靶向组、非靶向组和竞争抑制组)的叶酸靶向性;采用MTT细胞存活实验验证各组(对照组、非靶向组和靶向组)的肿瘤靶向治疗作用;采用细胞生长抑制实验对比不含DOX空白胶束和包含DOX的folate-PEG-PLGA胶束的细胞生长曲线,证明胶束自身安全性,并验证其内包抗癌药物的细胞毒性作用。结果荧光实验中,高表达的叶酸受体促进了细胞对folate-PEG-PLGA靶向胶束的摄取,靶向组的荧光强度大于非叶酸靶向组和竞争性抑制组;流式细胞仪进一步定量观察荧光强度,靶向组的细胞荧光强度[(89.26±1.19)%]显著强于非靶向组[(39.08±4.91)%,P=0.002]和竞争性抑制组[(26.86±6.5)%,P=0.005];MTT实验中,靶向组的细胞存活率显著低于非靶向组。结论 folate-PEG-PLGA胶束表面的叶酸配体和细胞膜表面的叶酸受体之间存在特异性靶向作用,这种靶向作用增加了肿瘤细胞对含药胶束的细胞摄取,增强了内包药物的细胞毒性和包药胶束的靶向治疗作用。
