Determination of Puerarin and Paeoniflorin in Zhangyanming Capsules and Tablets by HPLC

目的:建立用高效液相色谱法测定障眼明胶囊和片剂中葛根素和芍药苷含量的方法。方法:采用Diamonsil-C_(18)色谱柱(250 mm×4.6 mm,5μm),流动相:乙腈-0.1%磷酸溶液(13∶87),柱温20℃,流速为1.0 ml·min^(-1),检测波长为230 nm和250 nm。结果:葛根素在49.98~374.85 ng范围内线性关系良好(r=0.997 8),平均回收率为98.6%和98.7%。芍药苷在182.14~1 366.05 ng范围内线性关系良好(r=0.999 9),平均回收率为99.5%和98.7%。结论:该法简便,可靠,准确,可作为测定障眼明胶囊和片剂中葛根素和芍药苷含量的方法。

Objective ： To establish the method for determining the content of puerarin and paeoniflorin in Zhangyanming capsules and tablets. Method： An HPLC method was used with a Diamonsil Cis （250 mm x4.6 mm,5 m） column at 20℃. The mobile phase consisted of acetonitrile-0.1% H3 PO4 solution （ 13：87 ） and the flow rate was 1.0 ml . min-1. The detection wavelength was 230nm and 250nm. Result： The calibration curve of puerarin was linear within the range of 49.98-374.85ng（ r = 0.997 8 ）. The average recovery was 98.6% and 98.7%. The calibration curve of paeoniflorin was linear within the range of 182.14-1 366.05ng（ r = 0. 999 9）. The average recovery was 99.5% and 98.7%. Conclusion： The method is simple, reliable and accurate in the content determination of pu-erarin and paeoniflorin in Zhangyanming capsules and tablets.
