摘要
目的建立同时测定阴炎净洗液中苦参碱和氧化苦参碱含量的反相高效液相色谱法。方法色谱柱为XB—NH:柱(150mm×416mm,5μm),流动相为乙腈一无水乙醇-3%磷酸(80:12:8),检测波长为220nm,流速为1mL/min,柱温为40℃。结果苦参碱进样量在0.2~3.0μg范围内与峰面积呈良好线性关系(r=0.9997),氧化苦参碱进样量在0.2~3.0μg范围内与峰面积呈良好线性关系(r=0.9998),苦参碱和氧化苦参碱平均加样回收率分别为99.51%和100.00%。结论该方法简便、准确、可靠,适用于阴炎净洗液中苦参碱和氧化苦参碱的常规分析和质量控制。
Objective To establish a simultaneous determination method of the matrine and oxymatrine contents in Yinyanjing Lotion by reversed phase high-performance liquid chromatography(RP-HPLC). Methods The reversed phase high-performance liquid chromatog- raphy(RP- HPLC) method was adopted with the XB- N H: column(150 mm×416 mm, 5 μm), the mobile phase was acetonitrile- anhydrous ethyl alcohol-3% phosphoric acid (80:12: 8),with the flow rate of 1.0 mL/min, the detection wavelength of 220 nm and column tem- perature of 40 ℃. Results The sample size of matrine within the range of 0. 2 - 3.0 μg showed the good linear relation with the peak area( r = 0. 999 7); the sample size of oxymatrine within the range of 0. 2- 3.0 μg showed the good linear relation with the peak area (r =0. 999 8). The average recovery rate was 99. 51% for matrine and 100. 00% for oxymatrine. Conclusion The method is simple, accurate, reliable and suitable for the routine analysis and the quality control of matrine and oxymatrine in Yinyanjing Lotion.
