摘要
以短密青霉(Penicillium brevicompactum)TK1-41#为出发菌株,采用理性化筛选方式,对麦考酚酸(MPA)产生菌进行诱变育种研究。根据MPA产生菌的特点,设计了一种新的筛选方式。即通过制霉菌素预处理,改变了MPA产生菌细胞膜的通透性,提高了它对诱变剂的敏感性。最后再经过紫外诱变和添加高浓度自身代谢产物的方法,对MPA产生菌进行复合处理,获得了高产突变株TK93#,其在1T发酵罐上的产量比出发菌株TK1-41#提高了163%。
The mycophenolic acid (MPA) producing strain, Penicillium brevicompactum, #TK1-41 , as the parent strain, was subjected to a composite breeding selection studies and using of nystatin as a pretreatment agent to modify the permeability of the cell membrane of the parent strain, resulting to increase the sensitivity of mutagents. By the treatment of UV and screening the high tolerance to final product (MPA), a high MPA producing mutant #FK93 was obtained. In 1 m3 tonnage fermentor, the titre of#TK93 was 163% higher than that of the orizinal strain, #TK1-41.
