HPLC-ELSD测定人参首乌胶囊中人参皂苷Rg1等8种人参皂苷类成分 被引量:10

Determination of Eight Ginsenosides in Renshenshouwu Capsules by HPLC-ELSD

摘要 目的建立同时测定人参首乌胶囊中8种人参皂苷类成分含量的高效液相色谱-蒸发光散射器检测法。方法 UltimateC18(4.6 mm×250 mm,5μm)色谱柱,流动相为乙腈-水,梯度洗脱,流速1.0 mL·min-1,蒸发光散射检测器漂移管温度100℃,气体流量2.5 L·min-1,增益2。结果人参皂苷Rg1、Re、Rb1、Rc、Rb2、Rb3、Rd、20(S)-Rg3分别在0.483~12.1、0.444~11.1、0.502~12.6、0.490~12.3、0.455~23.2、0.464~11.6、0.264~13.2和0.244~12.2μg内成良好的线性关系(r>0.999 5),平均加样回收率分别为101.9%、104.5%、98.3%、97.9%、97.8%、101.2%、100.6%和99.0%。结论该方法简便、准确、分离效果好,无干扰,可用于人参首乌胶囊的质量评价。 OBJECTIVE To develop an HPLC-ELSD method for simultaneous determination of the contents of eight ginsenosides in Renshenshouwu capsules (traditional Chinese medicines ). METHODS The analysis was performed on an Uhimate C18 colunm (4. 6 mmx 250 ram,5 μm) eluted with mobile phase consisting of acetonitrile and water in gradient mode at a flow rate of 1.0 mL min-1 The column temperature was set at 30 C. The ELSD drift tube temperature was set at 100 C ,and the gas flow rate was set at 2.5 L min -1. RESULTS Good linearity(r 〉0. 999 5) was obtained in the range of 0. 483 -12.1 p,g,0.444-11.1 p,g,0.502-12.6 p,g,0. 490 - 12. 3 p,g,0. 455 -23.2 p^g,0. 464 - 11.6 p,g,0. 264 - 13.2 p^g and 0. 244 - 12. 2 p^g for ginsenoside Rgl ,ginsenoside Re ,gin- senoside Rbl,ginsenoside Rc,ginsenoside Rb2,ginsenoside Rb3,ginsenoside Rd and 20(S)- ginsenoside Rg3,respectively. The average recoveries of the eight compounds were 101.9% ,104. 5% ,98. 3% ,97. 9% ,97. 8% ,101.2% ,100. 6% and 99. 0 % ,respectively. CON- CLUSION The method is reliable, simple ,precise, and can be used for the quality control of Renshenshouwu capsules.
