Per2基因可增强人宫颈癌对射线的敏感性

Study on the effects of Per2 gene on enhancing radio sensitivity of human cervical cancer

摘要 目的:评价Period2(Per2)基因转染后过表达对人宫颈癌细胞(Hela)X射线放射敏感性的影响。方法:采用脂质体介导方法将Per2基因转染至Hela细胞,以空白组及空载体质粒pcDNA3.1(+)为对照,采用Wsetern Blot检测Per2基因在Hela细胞中的表达,给予X射线单次照射后,流式细胞仪检测细胞凋亡,采用细胞集落形成实验检测细胞增殖情况。结果:X射线照射后,与转染空载体质粒组及空白组比较,转染Per2基因Hela细胞凋亡率增加,细胞克隆形成减少。结论:节律基因Per2过表达使X射线照射的人宫颈癌Hela细胞凋亡增加,细胞增殖减弱,明显增强了HeLa细胞对放射损伤的敏感性。 Objective: To study the effects of Per2 gene over - expression on radio sensitivity of Hela cells . Methods: Lipo- feetamine mediated gene transfection method was used to transfect Per2 into Hela cells, blank group and empty plasmid pcDNA3 . 1 ( + ) as control. The expression of Per2 was determined by Wsetem Blot. After X - ray radiation, the apoptosis were analysed by FCM. The colony forming assay was used to show the cell proliferation . Results: Compared with control, transfected Hela cells revealed such characteristics after X - ray radiation as followed: increasing the rateof apoptosis, decreasing colony formation on plate. Conclusion : Circadian Per2 gene over - expression in Hela cells can increase the rate of apoptosis, decrease the cell proliferation, as a result, increase the radio sensitivity of this cell line to radiation damage .
