金欣口服液含药血清及其有效单体白藜芦醇对呼吸道合胞病毒感染Hep-2细胞白介素-6/白介素-8表达的调控作用

Study on effects of Jinxin Oral Liquid medicated serum and effective monomers resveratrol on the expression of IL-6 and IL-8 by Hep-2 cells infected with RSV

目的:探讨金欣口服液含药血清及其有效单体白藜芦醇(R)对呼吸道合胞病毒(RSV)感染人喉癌(Hep-2)细胞后炎症因子白介素-6(IL-6)和白介素-8(IL-8)表达的调控作用。方法:体外培养Hep-2细胞,A、B组分别行含药血清、单体干预,均以利巴韦林作为阳性对照药物,收集上清液检测。结果:RSV感染24h后,A、B两组中RSV组IL-6、IL-8表达量均显著增高,金欣组的IL-6、IL-8的表达量较RSV组有所下降,R组50μmol/L组能显著下调IL-6和IL-8的高表达,R组IL-6和IL-8的含量也有所下降,但无统计学意义。结论:金欣口服液含药血清及其有效单体R能够下调RSV感染Hep-2细胞IL-6和IL-8促炎性因子的高表达,推测其为抗RSV感染的作用机制之一,为临床上早期用药减轻炎症反应及研制、开发抗RSV的有效药物提供科学依据。

Objective： To investigate the regulation of Jinxin Oral Liquid medicated serum and effective monomers resveratrol on the expression of IL-6 and IL-8 by Hep-2 cells infected with RSV. Methods： In vitro experiment, the group A was intervened with medicated serum but the group B with monomers, then set Ribaviran positive control, the cell snperuatants collected after 24h storage. Results： The expression of IL-6 and IL-8 were up-regulated significantly compared with the control group after hep-2 cells infected, and they were down-regulated in Jinxin Oral Liquid groups compared with RSV infected group. Resveratrol in the dosage of 50pmol/L can typically down-regulate the expression of IL-6 and IL-8, which effect was the same as the dosage of 25pmol/L. Conclusion： Jinxin Oral Liquid medicated serum and its effective monomers resveratrol can down- regulated the expression of IL-6/IL-8 compared with RSV infected groups, which indicates that one of the mechanisms is anti- RSV infected. One scientifc basis is provided which reduced the inflammatory response of taking drugs early and the development of effective anti-RSV drugs.
