四妙散干预兔膝关节创伤性滑膜炎白细胞介素-1β和肿瘤坏死因子-α水平的研究 被引量:19

摘要 目的:通过创建兔膝关节创伤性滑膜炎动物模型,应用中药四妙散对其白细胞介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)进行干预,并观察组织形态学改变。方法:将24只家兔随机分为3组,空白组8只,模型组8只,受试组8只。从造模开始的第1天起,空白组及模型组均予以0.9%氯化钠溶液灌服,受试组予以四妙散(药液浓度为1g/mL)灌服,给药剂量为4.7mL/kg,2次/d,共10d。至第11天观察关节周径改变、关节体表温度变化,随后处死动物提取关节液,用酶联免疫吸附测定法(ELISA)检测各组兔膝关节液中IL-1β和TNF-α水平情况。取滑膜组织观察滑膜组织的病理切片。结果:空白组关节周径及体表温度低于模型组和受试组(P<0.01)。空白组IL-1β及TNF-α低于模型组和受试组(P<0.01);受试组IL-1β及TNF-α低于模型组(P<0.01)。空白组滑膜组织病理切片炎症积分低于模型组和受试组(P<0.05),受试组该积分低于模型组(P<0.05)。结论:①四妙散对治疗兔创伤性滑膜炎有较好的作用;②四妙散可以抑制IL-1β和TNF-α水平,从而发挥对创伤性滑膜炎的治疗作用。 Objective: To create the rabbit knee joint traumatic synovitis in animal models, application of Si Miao San to intervent whose levels of interleukin 113 (IL-1β) and tumor necrosis factor alpha (TNFa), and to observe the morphological changes. Methods: 24 rabbits were randomly divided into three groups, control group, model group and the test group, 8rats in each group. Started from the first day, the control group and model group were treated with saline gavage, and the test group was treated with Si Miao San (liquid concentration for lg/mL), gavage dose 4.7mL/kg, 2 times/d, a total of 10d. To 11 days observed joint circumference changes, joint surface temperature changes, and then animals were sacrificed to extract the synovial fluid, using enzyme-linked immunosorbent assay (ELISA) to detect the rabbit knee joint fluid IL-1β and TNF-u levels. Taken the synovial tissue and observe the synovial tissue biopsy to explore inflammation the expression levels of cytokines in knee joint synovitis in the pathological process and its cytological changes. Results: The joint circumference and temperature of the articular surface of control group were lower than model group and test group (P〈0.01). IL- 1β and TNF-a of control group were lower than test group (P〈0.01); IL-1β and TNF-a of test group were lower than model group (P〈0.01). The rabbit knee joint biopsy results showed that the inflammatory points of control group were lower than model group and test group (P〈0.05), and the control group was lower than model group (P〈0.05). Conclusion: ①Si Miao San have a better therapeutic effect on the rabbit knee joint traumatic synovitis; ②Si Miao San can inhibit IL- 1β and TNF-a levels, which play the role of the treatment of traumatic svnovitis.
