西黄丸通过ERK/MAPK信号通路对人结肠癌裸鼠移植瘤的影响 被引量:32

Effects of Xihuang Pill on the growth of human colorectal cancer cell xenografts in nude mice through the ERK/MAPK pathway

摘要 目的:探讨西黄丸通过ERK/MAPK信号通路对人结肠癌裸鼠移植瘤的影响。方法:建立人结肠癌裸鼠皮下移植瘤模型,24只实验裸鼠随机分为4组:对照组、西黄丸高、中、低剂量组;观察西黄丸对裸鼠致瘤的影响,计算肿瘤体积和移植瘤抑瘤率,绘制肿瘤生长曲线。光镜下观察瘤体、肺、肝、淋巴结等组织形态学改变,免疫组化法检测增殖细胞p-ERK1/2蛋白表达。结果:西黄丸可抑制移植瘤生长,并且可以降低p-ERK1/2蛋白表达。结论:西黄丸对人结肠癌裸鼠移植瘤有明显的治疗作用,延缓肿瘤的生长,抑制p-ERK1/2蛋白表达,可能与ERK/MAPK信号通路有关。 Objective: To investigate the effects of Xihuang Pill on the human colorectal cancer cell xenografts in nude mice through the ERK/MAPK signal transduction pathway. Methods: The nude mice model with human colorectal cancer cell was established and 24 mice randomly divided into four groups: including the control group and treatment groups ( high, medium, and low dosage group). During the experience, the effect of Xihuang Pill on the nude mice with tumor was examined, which included the tumor volume, inhibition rate and the growth curves of xenograft tumor. Histopathological examination of the tumor tissue, lung, liver and lymph node of nude mice were observed under optical microscope. The protein expression level of p-ERK1/2 was also detected by immunohistochemical assay. Results: Compared with the control group, the growth of the xenografts and tumorigenesis of human colorectal cancer were inhibited significantly by Xihuang Pill. In addition, the expression of p-ERK1/2 was reduced markedly after treatment. Conclusion: Xihuang Pill could inhibit the growth of xenografts when reduce the protein expression of p-ERK1/2. These data suggested that one possible mechanism may involve the regulation of ERK/MAPK signal transduction pathway.
