下瘀血汤全药与组分处方对免疫性肝纤维化大鼠肝组织α-SMA、TGF-β1、PDGF-βR mRNA表达影响的比较研究 被引量:4

摘要 目的:比较下瘀血汤全药与组分处方对免疫性肝纤维化大鼠肝组织α-SMA、TGF-β1、PDGF-βRmRNA表达的作用差异,探讨下瘀血汤全药与组分处方抗肝纤维化的作用机制。方法:Wistar大鼠随机分为正常组、造模组,造模组大鼠腹腔注射猪血清复制肝纤维化模型,每次0.5 mL/只,每周2次,共12周。造模成功后造模组大鼠随机分为模型组、下瘀血汤组、组分处方组,第8周开始下瘀血汤组、组分处方组大鼠造模同时口服给药,按10 mL·kg-1容积,下瘀血汤组每日2.34 g·kg-1,组分处方组每日0.648 g·kg-1,连续4周。正常组、模型组大鼠给与同体积生理盐水。12周末,处死动物,获取标本,realtime-PCR检测肝组织α-SMA、TGF-β1、PDGF-βRmRNA表达。结果:与正常组相比,模型组α-SMA、TGF-β1、PDGF-βRmRNA的表达均显著升高(P<0.05);与模型组相比,下瘀血汤组、组分处方组α-SMA、TGF-β1、PDGF-βRmRNA的表达明显降低(P<0.05);与组分处方组比较,下瘀血汤组α-SMA、TGF-β1、PDGF-βRmRNA表达明显降低(P<0.05)。相关性分析表明α-SMA、TGF-β1、PDGF-βRmRNA表达呈正相关。结论:下瘀血汤全药与组分处方可通过抑制α-SMA、TGF-β1、PDGF-βRmRNA表达而发挥抗肝纤维化作用,且下瘀血汤全药处方作用优于组分处方。 Objective : To investigate action mechanism of Xiayuxue Decoction and its ingredients recipe against liver fi- brosis, through comparing the difference of Xiayuxue Decoction and its ingredients recipe on α -SMA ,TGF - β1 and PDGF - βR mRNA expressions in porcine serum induced rats' fibrotic liver tissue. Methods. Wistar rats were randomly divided into normal group, model group and rat liver fibrosis modal was duplicated by intraperitoneal injection of porcine serum. After successful modeling,rats were randomly divided into model group, Xiayuxue Decoction group and its ingredients recipe group. Rats were given TCM preparation for 4 weeks. At the end of 12 weeks, animals were sacrificed to obtain liver tissue samples for determination of α - SMA, TGF - β1 and PDGF - 15R mRNA expressions. Results:Compared with normal group,α-SMA,TGF- β1 and PDGF- 15R mRNA expressions in model group were significantly increased ( P 〈0. 05) Compared with model group,or -SMA,TGF - β1 and PDGF - 15R mRNA expressions were significantly decreased in Xi- ayuxue Decoction and its ingredients recipe groups ( P 〈 0. 05 ). Compared with its ingredients recipe group, α- SMA, TGF -β1 and PDGF- BR mRNA expressions in Xiayuxue Decoction were significantly lowered( P 〈0. 05). Correlation analy- sis showed that α -SMA,TGF - β1 and PDGF - 13R mRNA expressions were positively correlated. Conclusion:The action mechanism of Xiayuxue Decoction and its ingredients recipe against liver fibrosis is to inhibit α - SMA,TGF - β1 and PDGF - 13R mRNA expressions, and Xiayuxue Decoction is superior to its ingredient recipe.
