摘要
选用乙醇二次回流法粗提莲籽通心粉中的莲心总碱,粗提物通过大孔树脂柱层析的吸附与洗脱进一步分离纯化。通过研究得出,D101大孔树脂较适宜进行莲心总碱的纯化,其最佳的吸附条件为:上样液浓度10.00mg/mL、pH值9、体积30mL(0.43BV)、流速 mL/min,此工艺得到最大吸附量为9.55mg/g;其最佳的洗脱条件为:乙醇浓度70%、pH值2、体积4BV、流速1.4mL/min,洗脱率达89.93%,经纯化后最终得率为64.39%。
In the paper, we extracted the alkaloids from lotus plumule powder by using the ethanol refluxing method, and used column chromatography to separate and purify alkaloids extracts further. The results showed that, D101 macroporous resin was more appropriate for purifying the alkaloids, and the maximum adsorbing capacity 9.55 mg/g was achieved with the following conditions: sample concentration 10.0 mg/mL, pH value 9, volume 30 mL (0.43 BV) and flow rate 1 mL/min. The best elution conditions were as follows: the ethanol concentration was 70%, with the pH value of 2, volume of 4 BV and flow rate of 1.4 mL/min, and the desorption ratio was up to 89.93%, while the final yield of the purified alkaloids was 64.39%.
