摘要
目的:探讨外周血及关节液中CD11c+CD123-髓系树突状细胞(Myeloid dendritic cell,mDC)的数量与初发类风湿关节炎(Rheumatoid arthritis,RA)的关系。方法:采集RA患者及对照组抗凝外周血、膝关节关节液,流式细胞术检测mDC的表达;将mDC数量与CCP、CRP、RF、ESR、DAS28、关节肿胀数和关节疼痛数进行相关性分析。结果:RA患者外周血中mDC的数量与对照组相比,差异具有统计学意义(P<0.05),外周血中mDC百分比与DAS28评分、CRP、ESR成负相关(P<0.05),与RF、CCP、关节疼痛数成正相关(P<0.05),与关节肿胀数无相关性。RA患者关节液中mDC的数量与对照组相比,差异具有统计学意义(P<0.05)。不同疾病活动度的RA患者外周血中mDC的数量差异有统计学意义。结论:初发RA患者体内mDC表达的变化,与免疫功能的紊乱有关,与疾病的发生发展有一定关系,这对进一步阐明RA的发病机制起到一定的积极作用。
Objective:To explore the relationship between the quantity of CD11c^+CD123^- myeloid dendritic cells in peripheral blood and synovial fluid with rheumatoid arthritis (RA). Methods : The anticoagulant peripheral blood and knee joint fluid of RA pa- tients and controls were collected. The percentage of CD11c^+CD123^- myeloid dendritic cells was detected by flow cytometry. The cor- relation between the quantity of CD11c^+CD123^- myeloid dendritic cells and the level of anti-CCP, CRP, RF, ESR, DAS28, the num- ber of tender ioints(TJ) and swollen joints(SJ) were analyzed. Results: Compared with the control group, the percent of the RA pa- tients' CD11c^+CD123^- myeloid cell in peripheral blood was statistically significant (P 〈 0. 05 ) ; compared the percentage of CD11c^+CD123^- myeloid dendritie cell in peripheral blood with DAS28 score, CRP, ESR , it showed a negative correlation (P 〈 0.05 ) ; com- pared the percentage of CD11c^+CD123^- myeloid dendritic cell in peripheral blood with RF, CCP, the number of TJ, it showed a posi- tive correlation; it was no correlation with the number of SJ. The number ofCD11c^+CD123^- myeloid dendritic cell in RA patients' joint fluid compared with the control group,it had a statistically significant (P 〈0. 05 ). The CD11c^+CD123^- myeloid cell number in peripheral blood were different between different disease activity of RA patients. Conclusion: CD11c^+CD123^- myeloid dendritic cell expression change in RA patients was related to immune dysfunction, which also had a certain relationship with the development of the disease, and it play a positive role in further elucidation of the pathogenesis of RA.
