CD147-shRNA重组质粒对裸鼠肝癌细胞SMMC-7721皮下移植瘤的抑制作用

摘要 目的研究CD147-shRNA重组质粒对裸鼠肝癌细胞SMMC-7721皮下移植瘤生长的抑制作用。方法将SMMC-7721细胞、转染空载质粒的SMMC-7721/pBS/U6以及转染本室前期构建的靶向肝癌细胞CD147的重组质粒的肝癌细胞SMMC-7721/pBS/U6/CD147-shRNA3接种到裸鼠皮下,观察移植肿瘤的生长情况。结果转染CD147-shRNA重组质粒的肝癌细胞移植瘤生长速度明显减慢;瘤块平均体积减小,重量减小,移植瘤生长明显减缓,抑瘤率为64.8%。结论转染CD147-shRNA重组质粒的SMMC-7721细胞致瘤性明显降低。 Objective To study the inhibition of recombinant plasmid CD147-shRNA on SMMC 7721 transplanta tion tumor in subcutaneous tissue of nude mice. Methods Vaccinate the SMMC 7721, SMMC-7721/ pBS/U6 and SMMC-7721/pBS/U6/CD147-shRNA3 into the subcutaneous tissue of nude mice,and observe the growth of the trans- plantation tumor. Results The transplantation tumor of transfected SMMC-7721 cells by CD147-shRNA grows signifi- cantly slowly, the volume and weight of the tumor is small. The growth of transplantation tumor is significantly inhibi- ted by the rate of 64.8%. Conclusion The tumorigenicity of transfected SMMC-7721 cell by recombinant CD147-shR- NA significantly reduced.
