大型溞N-乙酰-β-D-氨基葡萄糖苷酶纯化产物的动力学特性 被引量:1

摘要 为制备免疫原以测量个体生长和繁殖过程中所释放的几丁质酶,从大型溞(Daphnia magna)体内提纯了N-乙酰-β-D-氨基葡萄糖苷酶(EC 3.2.1.52),经硫酸铵沉淀分级分离、DEAE Sephadex A-25阴离子交换柱层析和Sephadex G-150分子筛柱层析纯化等步骤,酶的纯化倍数为20.09。纯酶比活力为2 243 380 U.mg-1。酶分子中各亚基的分子量分别为128,99和71 Ku。根据测试,酶促反应的米氏常数为0.300 mmol.L-1,最大反应速度为0.072μmol.mg-1.min-1。酶催化底物分解的最适pH为7.0,最适反应温度为45℃;酶在值4.2~7.8的范围内活性处于高位;在不高于35℃的温度下处理30 min,酶活性下降不明显。至于从大型溞培养液中获得的NAGase,其催化底物分解的最适pH值为7.4,最适反应温度为40℃,该酶在pH值4.6~7.8的范围内活性处于高位,在不高于40℃下处理30 min,酶活性下降不明显。这显示被提纯的酶和培养液中分离的酶在温度、pH以及热稳定性和酸碱稳定性方面均具有较高程度的相似性,有可能属于相同分子型。
