摘要
目的:分析和比较高脂饲料诱导Wistar和SD两种品系大鼠胰岛素抵抗模型的异同,为此类模型的建立提供参考及药物的研发提供动物实验平台。方法:Wistar大鼠和SD大鼠,随机分为Wistar N组(全价普通颗粒饲料),Wistar HFD组(合成高脂饲料),SD N组,SD HFD组。模型组给予高脂饲料喂养制备胰岛素抵抗模型,空白组给予普通饲料,连续喂养8 w。每周大鼠称重1次。实验结束后,检测血清中GLU、INS、CHO、TG、LDL-C水平,计算ISI、IRI。结果:8 w后,Wistar大鼠和SD大鼠均表现出糖耐量减低、血清INS水平升高、ISI降低、IRI升高,且伴有明显的血清脂质代谢紊乱,表明两种品系大鼠经高脂饲料喂养8 w后均能形成显著的胰岛素抵抗模型。但Wistar大鼠糖耐量降低、IRI增加较SD大鼠明显(P<0.05,P<0.01)。结论:高脂饲料喂养两种品系大鼠均可诱导形成胰岛素抵抗模型,Wistar大鼠较SD大鼠更易诱发形成胰岛素抵抗模型。
Objective :To analyze differences between insulin resistance SD rat models and insulin resistance Wistar rat models which were induced by high fat diet, so as to provide a reference for establishment of model and provide experi- mental platforms for drug development. Methods :Wistar rats and SD rats were respectively divided into Wistar N group (which were fed by full price ordinary granulated feed),Wistar HFD group(which were fed by synthetic high fat feed),SD N group, SD HFD group.Rats were weighed once a week. Then serum GLU, INS, CHO, TG, and LDL levels of rats were de- tected, ISI and IRI were calculated. Results :Insulin resistance SD rat models and insulin resistance Wistar rat models showed impaired glucose tolerance and significant lipid metabolic disorders, ascent of serum INS level and IRI, descent of ISI. But compared with SD rats, glucose tolerance of Wistar rats was decreased and their IRI was significantly increased (P〈0.05 ,P〈0.01 ). Conclusion:Insulin resistance Wistar rat models were more easily prepared than SD rat models.
