摘要 以转基因741杨杂种未成熟胚为外植体,对未成熟胚诱导萌发与不定芽再生,以及芽的继代与生根培养进行了研究并获得再生植株。结果表明,最适宜未成熟胚诱导萌发的培养基为MS+6-BA0.1mg·L。+NAA0.01mg.L-1+3%蔗糖,诱导萌发率最高可迭76.5%;未成熟胚离体培养最适取胚时间为胚龄20~27d;不同杂交组合未成熟胚诱导萌发率存在明显差异,最高的杂交组合为转基因株系pblx84K,诱导萌发率为66.1%。不定芽继代培养基为MS+6-BA(0.2—1.0mg/L)+IBA(0.05~0.3mg/L);生根培养基为1/2Ms+(0.2~0.5mg/L)IBA+1.5%蔗糖.生根率在95%以上。 We studied the immature embryo induced germination, adventitious bud regeneration, bud subculture and rooting culture by using immature embryos of transgenic 741 poplar hybrids as explants, then obtained the regenerated plants. The immature embryos get a highest induced germination rate of 76.5% when adding 6-BA (0.1 mg/L), 0.01 mg NAA and sucrose (30 g/L) in medium MS. The appropriate period to take embryo is from embryo age of 20-27 days until fully mature. There are obvious differences of immature embryo induced germination rate in different hybrid combinations, and the transgenic lines pbl x84K have the highest induced germination rate of 66.1%. The ingredients of adventitious buds subculture medium are 6-BA (0.2-1.0 mg/L) and IBA (0.05-0.3 mg/L) in medium MS. The ingredients of root medium are IBA (0.2-0.5 rag/L) and sucrose ( 1.5% ) in medium 1/2MS, and the rooting rate is more than 95%.
