CO_2对人宫颈癌Hela细胞VEGF-C和MMP9表达的影响

EFFECTS OF CARBON DIOXIDE ON THE EXPRESSION OF VEGF-C AND MMP9 IN HUMAN CERVICAL CANCER CELL LINE HELA

摘要 目的探讨体外模拟二氧化碳气腹对宫颈恶性肿瘤细胞生长转移能力的影响。方法体外培养人宫颈癌Hela细胞株,以CO2分别处理1,2,3h后,置于常规培养箱中分别培养24,48,72h,采用实时荧光定量PCR法及免疫细胞化学法检测各组Hela细胞VEGF-C、MMP9 mRNA和蛋白的表达。结果 CO2处理后Hela细胞VEGF-C、MMP9 mRNA和蛋白的表达均显著高于对照组(P<0.01);在不同CO2处理时间及培养时间下,VEGF-C、MMP9 mRNA和蛋白的表达均有差异(P<0.01)。结论 CO2环境可促进Hela细胞生长转移因子VEGF-C、MMP9表达增加,从而增强Hela细胞的生长转移能力。 Objective To explore the effects of carbon dioxide pneumoperitoneum on the growth and me- tastatic ability of cervical cancer cell line. Methods Human cervical cancer cell line Hela was treated with carbon dioxide for 1, 2, 3h, respectively, then placed in normal incubator for 24, 48, 72h. The expression level of mRNA of VEGF- C and MMP9 in these cells were examined by real- time PCR and immunocyto- chemistry. Results The mRNA expression of VEGF - C and MMP9 was significantly higher compared with controlled groups after the treatment of carbon dioxide(P^0.01). After the different treatment time of carbon dioxide and different incubation time, the mRNA expression of VEGF- C and MMP9 had signifi- cant changes(P^0.01). Conclusion Carbon dioxide can promote the expression of VEGF- C and MMP9, thereby enhancing the growth and metastatic ability of Hela cell.
