骨碎补水提液对大鼠成骨细胞分泌IGF-I的影响 被引量:2

摘要 本试验采用荧光定量PCR法观察含不同浓度骨碎补水提液的培养液对体外培养的大鼠第三代成骨细胞分别作用24h、48h、72h后对其胰岛素样生长因子(IGF-I)基因表达的影响。作用24h后,1×10-2 mg/mL、1×10-3 mg/mL、1×10-4mg/mL和1×10-5 mg/mL的IGF-I表达量极显著增加(P<0.01),1×10-1 mg/mL的IGF-I表达量显著增加(P<0.05)。48h后,1×10-3 mg/mL、1×10-4 mg/mL和1×10-5 mg/mL表达量极显著增加(P<0.01),1×10-2 mg/mL的IGF-I表达量显著增加(P<0.05)。72h后,各试验组均抑制IGF-I的表达。提示一定浓度的骨碎补水提液在成骨细胞早期具有促进IGF-I表达的作用。 Different concentrations of Rhizoma Drynariae were added to third-passage rat osteoblasts. The levels of IGF-I in rat OB were measured by FQ-PCR. Different concentrations of Rhizoma Drynariae aqueous extracts were added to rat OB. 24 hours after exposure, the expression of IGF-I genes was promo- ted in the groups of 1X 10-2mg/mL 1X 10-3mg/mL 1 X 10-4mg/mL 1X 10-smg/mL (P^0.01)and 1X 10-1mg/mL(P%0.05). After exposured 48h, the expression of IGF-I genes were also promoted in the groups of 1)K10-3mg/mL 1X10-4mg/mL 1X10-Smg/mL(P^0.01) and 1X 10-2mg/mL(P^0: 05). 72 hours after exposure, the expression of IGF-I genes was inhibited in all of the groups. These results indi- cated that certain levels of Rhizoma Drynariae aqueous extracts could promote the expression of IGF-I genes in the early period of osteoblasts.
