骨形态发生蛋白受体-ⅠB和-Ⅱ在人脑胶质瘤中表达及意义 被引量:2

Expressions of BMPR-ⅠB and BMPR-Ⅱand their significance in human glioma

摘要 目的探讨骨形态发生蛋白受体(bonemorphogenetieproteinreceptor,BMPR)-ⅠB和BMPR11在人脑胶质瘤发生、发展中的作用及其与临床病理分级间关系。方法20例正常脑组织(对照组)和40例不同级别人脑胶质瘤组织(胶质瘤组),采用RT-PCR技术检测BMPR-ⅠB、BMPR-ⅡmRNA表达水平,采用免疫组织化学法检测其蛋白质表达阳性率。结果胶质瘤组BMPRⅠBmRNA(0.332±0.102)、BMPR-ⅡmRNA(0.307±0.115)及其蛋白阳性率(32.50%、22.50%)较对照组(0.413±0.096、0.493±0.107、70.00%、60.00%)明显降低(P〈0.05);Ⅲ-Ⅳ级胶质瘤低于Ⅰ-Ⅱ级(P〈0.05)。结论BMPR-ⅠB、BMPR-Ⅱ表达与人脑胶质瘤的发生、发展密切相关,可能成为人脑胶质瘤分子诊断的指标和治疗靶点。 Objective To detect the role of bone morphogenetic protein receptor (BMPR)-ⅠB and BMPR-Ⅱ in the occurrence and development of human glioma, and the relationship with clinicopathological grading. Methods The mRNA expressions and protein positive rates of BMPR- ⅠB and BMPR-Ⅱ were detected in 20 samples of normal brain tissue and 40 samples of human glioma by using reverse transcription PCR and immunohistochemical methods. Results The mRNA expressions of BMPR ⅠB and BMPR-Ⅱ were 0. 332±0. 102 and 0. 307±0. 115, and their protein positive rate were 32.50% and 22.50% in human glioma, significantly lower than those in the normal brain tissues (0. 413± 0.096, 0.493±0. 107, 70. 00%, 60. 00%) (P〈0.05). The expressions of BMPR ⅠB and BMPR-Ⅱ mRNA were significantly lower in stage Ⅲ and Ⅳ than those in stage Ⅰ and Ⅱ (P〈0.05). Conclusion The expressions of BMPR- ⅠB and BMPR-Ⅱ are closely correlated with the occurrence and development of human glioma, and maybe the indexes for the molecular diagnosis and the treatment targets of human gliomas.
