摘要
从分子水平上发展高特异和高灵敏的分子识别探针对肝癌的特异性识别和早期诊断具有非常重要的意义.本文以人肝细胞性肝癌细胞株Bel-7404为靶细胞株,人胆管细胞性肝癌细胞株QBC-939为对照细胞株,利用Cell-SELEX技术筛选得到4条与肝细胞性肝癌细胞株高亲和识别的核酸适配体探针,与靶细胞的结合解离常数均在纳摩尔级水平.通过与不同肝癌及其他肿瘤细胞系结合分析表明,筛选的核酸适配体不但能区分其他类型的肿瘤细胞株,还能区分肝细胞性肝癌的其他细胞株,表现出对靶细胞株的高度特异的识别性能,为肝细胞性肝癌的特异识别、早期诊断和肝癌的病理分型提供了一种有效的手段和方法.
The development of molecular probes for Hepatic Carcinoma recognition with high sensitivity and specificity would be of great importance for the early diagnosis of liver cancer. In this work, with a cholangiocarcinoma cell line QBC-939 as counter cell, a hepatocellular carcinoma cell line Bel-7404 was used as target cells for the selection of aptamers based on Cell-SELEX. Four aptamers were selected which all could bind to target cells with an equilibrium dissociation constant (Kd) in the nanomolar range. Furthermore, these aptamers could recognize the target Bel-7404 cells with high specificity. These aptamers would serve as effective tools for early diagnosis of hepatocellular carcinoma and liver pathological classification.
