摘要
目的研究黄芩、大黄、五倍子、蛇床子4种药物及其配伍组合的中药复方对牙龈卟啉单胞菌(Porphyromonas gingivalis,Pg)生成内毒素脂多糖(lipopolysaccharide,LPS)和细菌超微结构的影响。方法采用鲎实验检测中药处理Pg后LPS含量,透射电镜观察中药处理后Pg形态结构的改变。结果黄芩、大黄、五倍子、蛇床子及其中药复方处理Pg后LPS生成量分别为(0.016 1±0.000 3)EU/mL、(0.020 1±0.000 6)EU/mL、(0.016 7±0.000 1)EU/mL、(0.024 0±0.000 0)EU/mL和(0.009 5±0.000 5)EU/mL,显著低于空白对照组的(0.027 1±0.000 0)EU/mL(P<0.05),中药复方组LPS含量显著低于4种单味药组(P<0.05),经中药复方处理后,Pg胞壁失去原有结构而明显变薄,细胞变形,大部分胞浆溶解消化、漏出呈空泡,少数变性,胞浆中可见黑色沉淀。结论中药复方可有效抑制Pg生成LPS,同时导致细菌结构改变。
Objective To evaluate the effects of Radix Scutellariae, Radix et Rhizoma Rhei, Galla ehinensis, Cnidium monnieri and their compound on lipopolvsaccharides (LPS)production and ultrastruetural changes in tile cell stnw.ture of Porphyromonas gingivalis (Pg). Methods Limulus test was adopted to detect production of LPS and transmission electron microscope was used to observe uhrastruetural changes in the cell structure of Pg. Results The amount of LPS production in Radix Scutellariae, Radix et Rhizoma Rhei, Galla chinensis, Cnidium momfieri and their compound group were (0.016 1 ±0.000 3) EU/mL, (0.020 1 ±0.000 6) EU/mL, (0.016 7±0.000 1) EU/mL, (0.024 0±0.000 0) EU/mL and (0. 009 5 ± 0. 000 5 ) EU/mL respectively, significantly lower than that in control group (0.027 1 ±0. 000 0) EU/mL (P〈0. 05 ). The Chinese medicine compound had significant lower amount of LPS than each single ingredient ( P 〈 0.05 ). Deformed cells and loss of their cellular integrity of Pg could be seen following treatment of Chinese medicine compound, such as thin cell wall, dissolved cytoplasm, vacuolar degeneration and black precipitations in the cytoplasm. Conclusion The Chinese medicine compound has good antibacterial effect on Pg.
