背景:胆管癌(CCA)预后较差,近来研究表明光动力疗法(PDT)在CCA治疗方面具有良好的应用前景。目的:研究二氢卟吩e6(Ce6)介导的PDT对人CCA细胞活性和白细胞介素(IL)-6表达的影响。方法:以不同强度光照和不同浓度Ce6干预人CCA细胞株QBC939,CCK-8法检测细胞活性,流式细胞术检测活性氧簇(ROS)含量,ELISA法检测细胞死亡,Annexin V-FITC/PI双染法检测细胞凋亡,ELISA法检测IL-6表达水平。结果:在相同Ce6浓度下,不同光照强度对QBC939细胞活性的影响无明显差异(P>0.05)。在相同光照强度下,QBC939细胞活性随Ce6浓度增加而逐渐降低(P<0.05),ROS含量、细胞死亡、细胞凋亡率、IL-6表达水平随Ce6浓度增加而显著升高(P<0.05)。结论:Ce6介导的PDT对人CCA细胞有显著杀伤作用,该作用与Ce6浓度具有量效关系。IL-6参与了PDT抗CCA的过程。
Background: The prognosis of patients with cholangiocarcinoma (CCA) is dismal. Recent studies indicated that photodynamic therapy (PDT) had a promising effect on CCA. Aims: To investigate the effect of chlorine e6 (Ce6)-mediated PDT on cytoactive and IL-6 expression of human CCA cells. Methods: Human CCA cell line QBC939 was treated with different intensity of illumination and different concentration of Ce6. Cytoactive was determined by CCK-8 assay. The content of reactive oxygen species (ROS) was determined by flow cytometry. Cell death was assessed by ELISA. Cell apoptosis was determined by Annexin V-FITC/PI double staining. The expression of IL-6 was determined by ELISA. Results: No significant difference was found in cytoactive detected when cells received same concentration of chlorin e6 and different intensity of photoradiation (P?0.05). Under the same intensity of illumination, cytoactive was significantly decreased and the content of ROS, cell death, apoptosis rate as well as the expression of IL-6 were significantly increased along with the concentration of Ce6 increased (P〈0.05). Conclusions: The Ce6-mediated PDT has significant killing effect on human CCA cells, and the effect appears to be correlated with the concentration of Ce6.
