摘要
目的目的选择一种准确、稳定、简便的西番莲果皮多糖含量测定的方法。方法分别用苯酚-硫酸法、蒽酮-硫酸法比色测定西番莲果皮中多糖的含量,比较2种方法的差异。结果苯酚-硫酸法、蒽酮-硫酸法分别在2.9~17.4μg/mL(r=0.999 4)和4.64~23.2μg/mL(r=0.999 3)范围内与吸光度呈良好的线性关系;精密度实验RSD分别为0.43%和0.55%;苯酚-硫酸法在2 h内稳定,蒽酮-硫酸法在1 h内稳定;含量测定结果分别为54.63 mg/g和48.19 mg/g。结论苯酚-硫酸法测定西番莲果皮多糖含量显色灵敏,颜色稳定,操作简便,可作为测定西番莲果皮多糖含量测定的方法。
Objective To select a more accurate, stable, and convenient method for the determination of Polysaccharide content in passiflora peel. Methods Polysaccharide content in passiflora peel was determined by 2 methods: phenol-sulfuric acid method and athranone-sulfuric acid method. Results The standard curve of phenol- sulfuric acid method and athranone-sulfuric acid method was respectively linear in the ranges of 2.9 -17.4 μg/mL (r = 0. 999 4 ) and 4.64 - 23.2 μg/mL ( r = 0. 999 3 ). The phenol-sulfuric acid method was stable within 2 h, and athranone-sulfuric acid method was stable within 1 h, the content was 54.63 mg/g and 48. 19 mg/g. Conclusion phenol-sulfuric acid method is sensitive, stable and convenient, and can be used for the determination of polysaccharide content in passiflora peel.
