摘要
目的 探讨硒对培养心肌细胞的抗氧化损伤作用。方法 应用不同浓度的过氧化氢 (H2 O2 ) ,分别作用不同时间 ,动态观察其对心肌细胞的损伤作用。同时观察硒对培养心肌细胞的保护作用。结果 H2 O2 作用于培养心肌细胞可造成乳酸脱氢酶 (LDH)从心肌细胞泄漏、细胞周期发生改变、心肌细胞和培养液中脂质过氧化物 (丙二醛 ,MDA)含量增加 ,同时发生 HE-染色可见的形态学改变。培养液中加入一定浓度的硒后 ,培养细胞和介质中 LDH和 MDA水平显著下降 (P<0 .0 1 ) ,噻唑蓝 (MTT)测定培养心肌细胞的存活率显著升高 ,细胞周期和形态学表现与正常细胞基本相同。结论 H2 O2 作为一种自由基 ,依其浓度和作用时间可造成不同程度的心肌细胞的损伤。硒作为具有抗氧化作用的微量元素 ,能明显抑制自由基对心肌细胞的损伤作用。
Objective:To evaluate the effect of Se on the oxidative injury in cardiomyocytes. [WT5HZ]Methods:[WT5BZ]Neonatal rat cardiomyocytes were cultured and their cytotoxicity induced by H 2O 2 over a wide concentration range(0.05 mmol/L 50 mmol/L)was dynamically assessed.Effect of Se on cardiomyocytes exposed to H 2O 2 was observed. [WT5HZ]Results:[WT5BZ]Application of H 2O 2 to cardiomyocytes caused LDH release,accumulation of lipid peroxide(MDA),and changes in cellular proliferation cycle,with concomitant morphologic changes.Supplementation of Se could decrease the release of LDH and MDA production in the cultured cardiomyocytes exposed to H 2O 2(P<0.01),and significantly increase the viability of cardiomyocytes by MTT test. [WT5HZ]Conclusion: [WT5BZ]H 2O 2 can induce cardiomyocyte cytotoxicity in a dose and time dependent manner.The magnitude of the cytotoxicity by exposure to H 2O 2 is reduced by Se supplementation.It is suggested that Se,as an antioxidant,may protect cardiomyocytes against reactive oxygen radicals.
