摘要
目的 探讨牛磺酸和复合微量营养素对光照和暗适应条件下大鼠视网膜代谢型谷氨酸受体 6亚型 (m Glu R6) m RNA的影响。方法 在幼年大鼠饲料中添加牛磺酸或 VA、VB1、VB2 、尼克酸、Zn、Se等复合微量营养素 ,营养干预 4wk后 ,采用原位分子杂交技术检测各组大鼠视网膜 m Glu R6m RNA在光照和暗适应条件下的分布和表达。结果 光照和暗适应时 m Glu R6m RNA均集中表达于内、外核层和神经节细胞层。牛碘酸能增加光照和暗适应时上述部位m Glu R6m RNA的表达 ,而复合微量营养素对其表达无明显影响。结论 除双极细胞外 ,部分神经节细胞也表达 m Glu R6;光感受器突触前膜存在反馈性的 m Glu R6。牛磺酸可能通过促进光照和暗适应条件下 m Glu R6m RNA的表达 。
Objective:To investigate the effect of taurine or micronutrient complex vitamin A,vitamin B 1,vitamin B 2,niacin,zinc and selenium on the expression of metabotropic glutamate receptor subtype 6(mGluR6)mRNA in rat retina in light and dark adaptation. [WT5HZ]Methods:[WT5BZ]After supplementation of taurine or micronutrient complex in rat diet for 4 weeks,the localization and expression of mGluR6 mRNA in rat retina in light and dark adaptation were observed with in situ hybridization. [WT5HZ]Results:[WT5BZ]mGluR6 mRNA was expressed in the outer nuclear layer(ONL),the inner nuclear layer(INL),and ganglion cells layer(GL)in light and dark adaptation.Taurine markedly increased the expression of mGluR6 mRNA in ligh and dark adaptation,but micronutrient complex had no effects on it. [WT5HZ]Conclusion:[WT5BZ]Execpt rod bipolar cells,mGluR6 is located on the presynaptic membrane of photoreceptors,and partially expressed on ganglion cells.Taurine can modulate rod visual signal transmission by affecting the expression of mGluR6 mRNA.
