摘要
目的 观察 P-糖蛋白 (P- gp)与蛋白激酶 C同工酶亚型 (PKCα)在 SGC790 1/ VCR耐药细胞系的相关表达及其功能 .方法 采用免疫细胞化学检测 P- gp在 SGC790 1/ VCR及其亲本药敏细胞 SGC790 1的表达 ;免疫荧光双标及激光共聚焦显微镜技术 (L CSM)观察 P- gp与 PKCα的相关表达 ;流式细胞仪检测细胞内罗丹明 12 3(R12 3)的荧光强度 .结果 P-gp及 PKCα在细胞膜上有共同表达部位 ,P- gp与 PKCα相关表达在 SGC790 1/ VCR 比 SGC790 1细胞明显增加 ;与SGC790 1相比 ,SGC790 1/ VCR细胞内罗丹明 12 3的荧光强度明显减弱 .结论 P- gp及 PKCα在 SGC790 1/ VCR耐药细胞系的耐药中起重要的作用 .
AIM To analyse the related expression and function of p-glycoprotein (P-gp) and protein kinase C (PKC) isoform in gastric cancer multidrug-resistant cells SGC7901/VCR. METHODS Immunohistochemical method was used to determine the expression of P-gp in SGC7901/VCR.The related expression of P-gp and PKCα was observed by double-labeled immunofluorescent and laser confocal scanning microscopy. The accumulation of rhodamine 123 in cells was determined by flow cytometry. RESULTS There was co-expression of P-gp and PKCα on cell membrane, and the co-expression levels in SGC7901/VCR were higher than those in SGC7901. The accumulation of rohdamine 123 significantly decreased in SGC7901/VCR, compared with SGC7901. CONCLUSION P-gp and PKCα play important roles in multidrug resistance of SGC7901/VCR.
