尖吻蝮蛇毒PCA在急性心肌梗死诊断中的应用 被引量:5

摘要 目的:探讨尖吻蝮蛇毒血浆蛋白C激活物(protein C activator,PCA)在急性心肌梗死诊断中的应用。方法:30只雄性SD大鼠随机分为3组(n=10),即对照组(C group)、假手术组(S group)和急性心肌梗死组(AMI group);通过大鼠左冠状动脉前降支结扎术制作急性心肌梗死模型,用Medlab生物信号采集系统检测心电变化。发色底物法测定血浆蛋白C的活性,酶联免疫吸附法(ELISA)检测血管内皮细胞蛋白C受体(EPCR)的含量,凝血仪检测活化部分凝血活酶时间(APTT)。结果:AMI组心电显示ST段弓背向上抬高,QRS波群形态改变,T波增高;与S组相比较,AMI组大鼠血浆蛋白C活性1 h即显著降低(P<0.05);2 h后EPCR含量明显减少(P<0.05);APTT明显缩短(P<0.05)。结论:以尖吻蝮蛇毒PCA制备的血浆蛋白C活性检测试剂可较灵敏地反映心肌梗死大鼠血浆蛋白C活性的变化。 Objective:To assess the protein C activator ( PCA) extracted from the venom of Agkistrodon acutus in diagnosis of acute myocardial in-farction( AMI).Methods:Thirty SD male rats were equally randomized into groups of controls ( C group ) , sham operation ( S group ) and AMI ( AMI group) .Animal models of AMI were developed through ligation of the left anterior descending of the coronary artery , and examined for the changes of electrocardiogram with Medlab bio-signal operating system. Chromogenic substrate assay was performed to detect the PC activities ,and enzyme-linked immunosorbent assay( ELISA) was carried out to measure the plasma content of endothelial cell protein C receptor(EPCR).Auto-matic coagulation analyzer was used to determine the activated partial thromboplastin time ( APTT ) .Results:The echocardiographic recording revealed that ST segment and T wave were significant elevated after induc-tion of AMI.Significantly decreased PCA activity at 1 h,reduced EPCR level at 2 h and shortened APTT were observed in AMI group as compared with S group(P〈0.05).Conclusion:PCA kit prepared from the venom of Agkistrodon acutus may sensitively indicate the changes of serum PC in rats with AMI.
