摘要
文章以杜泊×小尾寒羊为研究对象,采用PCR-SSCP方法对H-FABP基因exon2进行SNP检测,结果发现,g.939 A→G、g.980 G→A、g.1013 A→C、g.1018 T→C四个突变。与部分肉质性状、脂肪酸、氨基酸相关分析发现,基因型BB对剪切力有显著负效应(P<0.05),基因型BB对棕榈酸含量有显著正效应(P<0.05),基因型BB型对硬脂酸、亚油酸含量有极显著负效应(P<0.01),基因型BB对谷氨酸、丝氨酸、苏氨酸、丙氨酸含量以及氨基酸总和均有显著负效应(P<0.05),对脯氨酸含量有极显著负效应(P<0.01)。
PCR-SSCP approach was applied to test the single nucleotide polymorphisms (SNPs) at the exon 2 of H-FABP (Heart fatty acid-binding protein) gene and DorperxSrnalI-Tailed Han sheep as the research object in this study. The results showed that four mutations (g.939 A→G,g.980 G→A,g.1013 A→C, g.1018T→C) were detected. The correlation analysis on polymorphisrns with part meat quality trait, fatty acids and amino acids showed that genotype BB had a negative effect on shear force(P〈0.05), genotype BB had a significantly positive effect on palrnitic acid content(P〈0.05), genotype BB had a significantly negativeeffect on steadc acid and linoleic acid content(P〈0.01), genotype BB had a certainly negative effect on glutamic acid, serine, threonine, alanine and the total amino acid(P〈0.05) and had a significantly negative effect on proline content(P〈0.01).
