摘要
目的 :探讨大蒜预防胃癌的细胞学机制 ,为大蒜预防胃癌提供理论依据。方法 :以低分化胃癌细胞系MGC - 80 3为靶细胞 ,用MTT法检测大蒜汁 (GJ)对肿瘤细胞活性的影响 ,透射电镜、DNA凝胶糖电泳和原位缺口标记法 (TUNEL)分析细胞凋亡。结果 :GJ能有效抑制MGC -80 3细胞的活性 ,并诱导其凋亡。形态学变化表现为细胞皱缩 ,染色质凝集、碎裂 ,沿核膜内缘分布 ,凋亡小体形成等 ;基因组DNA沿核小体之间断裂 ,电泳得到凋亡特征性的梯形条带 ;TUNEL法分析显示 ,GJ诱导的凋亡率呈药物浓度和作用时间依赖性。结论 :GJ能有效诱导胃癌细胞凋亡 ,是其预防胃癌的重要机制之一。
Objective:To explore the cytological mechanisn of actions of garlic juice(GJ) preventing human gastric cancer. Methods: The cytotoxicity rate of gastric cancer cells(MGC-803) was studied by MTT assay,and apoptosis of MGC-803 cells was analyzed by transmission electron microscopy, DNA agarose gel electrophoresis and TUNEL method. Results:GJ had a remarkable cytotoxic effect on gastric cancer cells.After 24 hours of exposure to GJ, gastric cancer cells presented some morphologic features of apoptosis, including cell shrinkage, nuclear condensation, DNA fragmentation and formation of apoptotic bodies. GJ induced apoptosis in MGC-803 cells in a dose-and time-dependent manner. Conclusion: The results suggested that the induction of apoptosis of cancer cells would be a very important mechanism of GJ preventing human gastric cancer.
