摘要 目的 :评价国产乙基纤维素微球的安全性及其在口腔颌面部动脉的栓塞效果。方法 :采用体外细胞培养技术 ,以 MTT法检测乙基纤维素微球的细胞生物学效应 ;在 X线监控下 ,将乙基纤维素微球选择栓塞在犬颈外动脉及其分支 ,在栓塞后的即刻和一月后测量其颈部各主要动脉的血液流速及流量 ,进行统计分析。不同时间取栓塞组织石蜡切片 ,常规 HE染色 ,光镜观察。结果 :细胞学实验表明 ,国产乙基纤维素微球的浸提液不影响细胞生长和增殖。动物实验表明 ,被栓塞血管的流量、流速明显降低 ,基本不形成侧枝循环 ,临近动脉的血流量则代偿性的增加。组织学观察可以见到微球成簇状堵塞末梢动脉血管 ,达到较为完全的栓塞效果。结论 :国产乙基纤维素微球生物相容性良好 ,可以持久阻塞动脉血管腔 ,有望在临床上用于血管性疾病的栓塞治疗。 ?Objective:To evaluate the safety and embolization effects of ethylcellulose microsphere(ECM) in oral and maxillofacial region. Methods:The in vitro cytotoxicity of ECM was tested by MTT assay;selective artery embolization was conducted by perfusion of ECM into internal maxillary artery of dogs under X ray monitor. Blood flow velocity and volume was measured at different time after embolization.All the data were analyzed statistically. The sections of embolized tissue at different time were examined microscopically. Results: There was no significant difference among the values of the groups in which L929 cells were treated with 1,1/2,1/4,1/8,1/16 and 1/32 of ECM extract for 1,3 and 5 days respectively. The blood flow of the embolized artery was completely blocked 1 h after the infusion of ECM without recovery in 1 month, while the blood flow volume of adjoining arteries increased compensatively. ECM were found to closely stuff the cavity of the artery from the histological examination. Conclusion: ECM can be used to embolize artery effectively without cytotoxicity.
