丹参注射液对小鼠心肌干细胞的影响 被引量:2

摘要 目的:探讨丹参注射液对小鼠原位心脏成体干细胞数量和旁分泌功能的影响。方法:以不同剂量丹参注射液对小鼠进行灌胃给药,分为高剂量丹参注射液组(5g·kg-1·d-1)和低剂量丹参注射液组(0.5g·kg-1·d-1),对照组采用相同体积生理盐水,每天1次连续1周。1周后经小鼠眼眶取血,检测血常规及肝肾功能;以小鼠新鲜心脏组织冰冻切片行免疫荧光染色,检测Sca-1+成体干细胞数量;以Real-Time PCR检测心脏血管内皮生长因子(VEGF)、IL-6、BAD基因表达情况。结果:灌胃1周后,三组间血常规、肝肾功能差异无统计学意义(P>0.05);与高剂量丹参注射液组比较,低剂量丹参注射液及对照组中Sca-1+成体干细胞数量及VEGF和IL-6基因表达显著减少,差异具有统计学意义(P<0.05)。同时,丹参注射液高剂量组BAD基因表达较丹参注射液低剂量组和对照组显著降低,差异具有统计学意义(P<0.05)。结论:丹参注射液能增多小鼠心脏中成体心脏干细胞数量,改善旁分泌功能。 AIM: To investigate the effect of Salviae miltiorrhizae on the number and parac fine effect of resident cardiac stem cell. METH- ODS: Mice were divided into three groups and received intragastric administration of high dose Salviae mihiorrhizae (5 g .kg -1.d-1 ), low dose Salviae miltiorrhizae (0.5 g. kg 1 -1 d-1) or saline for one week. Blood was collected for analysis of blood routin test, liver and renal function. The number of resident Sea-1+ cardiac stern cells was detected by immunofluorescent staining and the expressions of VEGF, IL-6 and BAD in heart tissue were determined with Real Time PCR . RESULTS: The number of white blood cell, red blood cell, platelet, liver and re nal function showed no difference among the three groups (P〉0.05), by intragastric admin-istration for 1 week. Compared with the highdose Salviae miltiorrhizae group, the number of resident Sca-1+ cardiac stem cells and the ex pression of VEGF, IL-6 were decreased in low dose Salviae miltiorrhizae group and control group, there was statistically significant differ ence (P〈0.05). Meanwhile, compared the oth er two groups , the expression of BAD was de-creased in the high dose Salviae miltiorrhizae group, there was statistically significant differ ence (P〈0.05). CONCLUSION: Salviae miltior rhizae treatment can increase resident Sca-1+ cardiac stem cells number and improve paracrine function.
