丙酮酸脱氢酶激酶1与鼻咽癌患者临床病理特征及预后关系 被引量:1

Association of PDK1 and clinicopathological features and prognosis in nasopharyngeal carcinoma

摘要 目的研究丙酮酸脱氢酶激酶1(pyruvate dehydrogenase kinase 1,PDK1)在鼻咽癌组织中的表达与临床病理因素的关系及其对预后的影响。方法应用免疫组化法检测52例鼻咽癌组织和20例鼻咽黏膜慢性炎症组织中PDK1蛋白的表达情况,统计分析PDK1表达与各临床病理特征以及其对鼻咽癌患者预后关系。结果PDK1在鼻咽癌组织中阳性表达率为61.5%(32/52),而鼻咽黏膜慢性炎症组织为阳性表达率为35.0%(7/20),不同组织PDK1蛋白表达差异有统计学意义(χ2=4.098,P<0.05)。PDK1蛋白的表达与颈部淋巴结转移及颅神经侵犯以及T分级相关,与患者性别、年龄、临床分期等无显著相关性。生存分析显示,PDK1蛋白阳性表达组患者的中位生存时间为51个月,阴性表达组为58个月。结论 PDK1基因可能在鼻咽癌的发生、发展、淋巴结转移和预后中起着重要作用,PDK1阳性表达可作为鼻咽癌不良预后因素。 OBJECTIVE To investigate the expressions of PDK1 gene and their correlation with the clinicopathological factors and prognosis in nasopharyngeal carcinoma(NPC).METHODS PDK1 expressions were detected by immunohistochemical staining in 52 specimens of NPC and 20 specimens of chronic nasopharyngitis tissues.RESULTS PDK1 expression was detected in 61.5% of the cancer tissues,significantly higher than that in nasopharyngitis tissues(35.0%,χ2=4.098,P 0.05).PDK1 expression was not correlated with gender,age,clinical stage.However,PDK1 expression was correlated with lymph node metastasis,cranial nerve palsy.Survival analysis showed that patients with high PDK1 expression had a shorter survival time and a lower 3 and 5 year survival rates than that in the patients with low expression.CONCLUSION PDK1 g e n e m a y p l a y a n i m p o r t a n t r o l e i n t h e carcinogenesis,cancer progression and lymph node metastasis in nasopharyngeal carcinoma.PDK1 gene may be an independent indicator of poor prognosis of nasopharyngeal carcinoma.
