维生素D3通过调节RUNX3抑制腹膜间皮细胞纤维化 被引量:1

摘要 目的探讨维生素D3对人腹膜间皮细胞(HPMC)纤维化的抑制作用及其可能机制。方法将永生化的人HPMC分为2组培养48 h,对照组为正常糖浓度组(葡萄糖浓度5.6 mmol/L),另一组为高糖处理组(葡萄糖浓度50 mmol/L);免疫组化检测HPMC中上皮细胞转分化过程中的上皮标志蛋白上皮型钙黏素(E-cadherin)和间皮细胞标志蛋白α-平滑肌肌动蛋白(α-SMA)的表达。用无水乙醇将维生素D稀释至(10-10、10-9、10-8)mol/L三种不同浓度后处理HPMC 24 h;Western blot法及实时定量PCR(qRT-PCR)检测RUNX3、E-cadherin及α-SMA的表达;构建重组质粒RUNX3-siRNA并转染高糖培养的HPMC,以获得敲减RUNX3基因的HPMC,Western blot法检测10-8mol/L维生素D3处理转染后各组细胞RUNX3、E-cadherin和α-SMA表达情况。结果高糖可诱导HPMC中E-cadherin蛋白水平显著下降,而α-SMA蛋白表达水平显著增加,提示高糖作用后HPMC发生上皮间质转分化;维生素D3可诱导高糖处理的HPMC中RUNX3 mRNA和蛋白表达水平增加,且这种作用存在剂量依从性。同时发现E-cadherin表达水平逐渐增加,α-SMA表达水平逐渐降低。以上结果提示:维生素D3可能通过调控RUNX3抑制高糖处理的HPMC发生上皮细胞转分化。敲减RUNX3基因后,维生素D3处理的高糖培养HPMC中E-cadherin及α-SMA表达水平无明显变化。结论维生素D3可能通过调控RUNX3的表达抑制HPMC的上皮间质转分化,从而抑制腹膜纤维化进程。
