摘要
为研究ANXA2表达水平对结直肠癌细胞(Caco2)形态学的影响,将ANXA2特异性干扰重组质粒pU6H1-GFP-siANXA2导入Caco2细胞,用扫描电子显微镜在转染后不同时间点对Caco2细胞的形态及表面结构进行观察.结果显示,抑制ANXA2表达后Caco2细胞的外观形态改变,细胞表面微绒毛及伪足等结构受损,并随时间延长而加剧,最终细胞表面趋于光滑,部分细胞出现凋亡表型.这些结果提示,ANXA2高表达是维持Caco2细胞活跃运动力有关恶性形态学特征的重要因素.
ANXA2 was excessively expressed in human colorectal carcinoma cells (Caco2). To study the effects of the excessive ANXA2 expression on surface morphology, the ANXA2 specific RNAi plasmid (pU6H1-GFP-siANXA2) was introduced into Caco2 cells. Then the cell surface morphology was observed using scanning electron microscope at different time point post transfection. The results showed that the cell shape was altered, meanwhile, the microvilli and pseudopodia structure were damaged when ANXA2 expression was inhibited. Moreover, the extent of the damage increased in a time dependent manner. At last, the cell surface became smooth and a part of cells appeared apoptotic changes. The results indicated that the excessive ANXA2 expression maintains the malignant surface morphology of Caco2 cells.
