摘要 目的 探讨bxl-xS基因对化疗后大肠癌细胞生物学行为的影响。方法 用基因转染方法进行LoVo细胞bcl-xS基因转染,然后用MTT和ELISA法检测bcl-xS转染细胞对化疗药MMC及5-FU敏感性的影响和作用机制,结果 bcl-xS转染可增强LoVO细胞对化疗药MMC及5-FU的敏感性,与经neo转染的细胞相比有显著差异(P〈0.05),ELISA细胞死亡试剂盒研究发现。 Objective To study the effect of bcl-xS gene transfection on the biological behavior of colorectal carcinoma cell. Methods We used a lipofectamine-mediated gene transfection technique to transfect LoVo cells with bcl-xS gene, the effect of which on the chemosensitivity of LoVo-bcl-xS gene-transfected cells to MCC and 5-FU were examined by MTT and ELISA methods. Results The results showed that bcl-xS gene transfection could significantly enhance the chemosensitivity of LoVo cells to MMC and 5-FU(P<0.05), evidenced by the comparison with neo-transfected LoVo cells. The findings by ELISA test with cell death examining kit indicated that the increased chemosensitivity to MMC and 5-FU was due to cellular apoptosis, and a 1 to 4 fold increase of the apopoptotic bcl-xS-transfecfed LoVo cells due to MMC and 5-FU treatment was observed, compared with that in LoVo-neo control group. Conclusion bcl-xS gene transfection can be of value in the gene therapy for colorectal malignancies.
