摘要
将HSV-I免疫过的小鼠淋巴结细胞在体外扩增,收集富集HSV-I抗原特异性的T细胞与BW 5147胸腺瘤细胞融合。对融合后产生的杂交瘤细胞进行抗原特异性及MHC识别限制选择。结果所得的T杂交瘤细胞具有在HSV-I特异刺激下,MHC一致时,有分泌IL-2的能力,并在体内耳廓病毒清除中发挥作用。它们是HSV-I抗原特异性,H-2~d限制的,具有辅助活性的T杂交瘤细胞。
Murine Tcell hybridoma were produced by fusing HSV-Ⅰ Specific enriched T cells with BW5147 thymoma cells. The hybrids were screened for their ability to produce IL-2 after exposure to Ag-pulsed, X-irradiated spleen cell monolayer of appropriate H-2 heplotype. These hybrids selected were able to produce HSV-1 specific IL-2, and showed the activity in ear virus clearance in vivo. they were identified HSV-Ⅰ specific, H-2d restricted T helper hybri-domas.
