摘要 目的:研究白介素 18( IL- 18)等内源性细胞因子对大肠癌细胞 FasL表达水平的调控作用。方法:采用 Western蛋白印迹法测定细胞因子处理后大肠癌细胞的 FasL表达水平。结果:在检测的 6株大肠癌细胞中,全部表达 FasL蛋白; TNF-α和 IFN-γ显著上调 DLD- 1 FasL蛋白的表达水平; TNF-α和 IFN-γ 以及乙酸肉豆佛波醇 (phorbol 12- myristate 13- acetate,PMA)加离子霉素 (ionomycin)显著上调 SW620的 FasL蛋白表达水平,并且 TNF-α和 PMA加离子霉素处理增加 SW620的 FasL蛋白表达具有时间效应;低浓度 IL- 18增强 DLD- 1和 SW620的 FasL蛋白表达,并具有时间效应。结论: TNF-α和 IFN-γ 以及 IL- 18等细胞因子具有上调大肠癌细胞 Fas配体表达的作用。 Objective: The current study was designed to investigate regulation effect of endogeneous cytokines to Fas ligand protein expression in human colon carcinoma cell lines. Method: Six human colon cancer cell lines were examined by using Western blot analysis for Fas ligand(FasL) expression and following treatment with TNF-α , IFN-γ and IL- 18. Results: Fas ligand protein was expressed in all of these six cancer cell lines. TNF-α and IFN-γ significantly upregulated the expression of FasL protein in DLD- 1. FasL expression in SW620 was upregulated by TNF-α , IFN-γ and PMA plus ionomycin, and by TNF- alpha and PMA plus ionomycin in a time- dependent fashion. FasL expression was also upregulated by interleukin- 18 at low concentration in DLD- 1 and SW620 in a time- dependent fashion. Conclusion: These data suggest that FasL expressed in human colon cancer cells may be regulated by endogenous cytokines such as interleukin- 18, TNF-α and IFN-γ .
