摘要
目的了解胃癌细胞SGC7901及其耐药细胞SGC7901/VCR中HSP90β蛋白的表达,与化疗药物敏感性的关系及降低细胞HSP90β蛋白的表达是否影响化疗药物的敏感性。方法用免疫组化法检测胃癌细胞SGC7901及其耐药细胞SGC7901/VCR中HSP90β蛋白的表达。MTT法测定SGC7901、SGC7901/VCR细胞及其相应的HSP90β蛋白低表达细胞AH-SGC7901和AH-SGC7901/VCR对化疗药物的敏感性,流式细胞仪测定细胞内药物含量。结果SGC7901/VCR细胞HSP90β蛋白的表达高于SGC7901细胞。SGC7901/VCR细胞对ADM、VCR、MMC和CTX的IC50分别为0.398、1.25、0.199和3 981.07μg/ml。SGC7901对ADM、VCR、MMC及CTX的IC50分别为1.58×10-6、1.99×10-3、3.1×10-2及630μg/ml。SGC7901/VCR相对于SGC7901细胞对ADM、VCR、MMC和CTX的耐药指数分别为2.52×105、6.28×102、6.42和6.32。
Objective To evaluate the expression of HSP90β in SGC7901 of human gastric cancer cell line and in SGC7901/VCR of MDR-type human gastric cancer cell line and its correlation with drug sensitivity of tumor cells and to study whether decreasing the expression of HSP90β could influence the tumor cell sensitivity to chemotherapeutic agents. Methods Immunohistochemical method was used to evaluate the expression of HSP90β in SGC7901 and SGC7901/VCR cell lines. AH-SGC7901 and AH-SGC7901/VCR cells were their corresponding low HSP90β expression cell lines. MTT was used to evaluate their sensitivity to Adriamycin ADR vincrinstine VCR mitomycin MMC and cyclophos-phamide CTX and flow cytometry was used to determine intracellular drug concentration of these cells. Results The expression of HSP90β protein in SGC7901/VCR cell line was higher than that in SGC7901 lines. IC50 of SGC7901/VCR cell to ADR VCR MMC and CTX were 0.398 1.25 0.1999 and 3981.07μg/ml respectively and IC50 of SGC7901 cell to ADR VCR MMC and CTX were 1.58×10-6 1.99×10-3 3.1×10-2 and 630ug/ml respectively. Drug resistance index of SGC7901/VCR to ADR VCR MMC and CTX as compared with SGC7901 were 2.52×105 6.28×102 6.24and 6.32 respectively and sensitivity of AH-SGC7901 and AH-SGC7901/VCR cells to chemotherapeutic drugs was much increased. The sensitivity of AH-SGC7901 to ADR VCR and MMC increased 1010000 and 24.8 times respectively as compared with its parent cell lines and its sensitivity to CTX had no obvious change. The sensitivity of AH-SGC7901/VCR to ADRVCRMMC and CTX incresed 3.98 6.28 3.15 and 5.01 times respectively as compared with its parent cell line. The mean fluorescence intensity of ADR in AH-SGC7901 and AH-SGC7901/VCR cell was significantly increased P< 0.05. Conclusion The expression of HSP90β protein in SGC7901/VCR cell line was higher than that in SGC7901 cell lines. The sensitivity of AH-SGC7901 and AH-SGC7901/VCR cells to chemotherapeutic drugs was much increased and the drug sensitivity of AH-SGC7901 cell was increased more obviously than that of AH-SGC7901/VCR cell.
