摘要
目的:探讨ras癌基因及p16抑癌基因与卵巢癌生物学行为的关系。方法:应用S-P免疫组化法及聚合酶链反应-单链构象多态性(PCR-SSCP)技术,检测36例卵巢癌,20例卵巢良性肿瘤及10例正常卵巢ras、p16基因及表达状况。结果:卵巢癌及对照组ras癌基因无突变及缺失。卵巢癌ras p21阳性表达80.6%,卵巢良性肿瘤仅有25%弱阳性表达,正常卵巢阴性表达,P< 0.01、P< 0.001,差异非常显著;卵巢癌及对照组p16基因无突变,卵巢癌p16缺失9例占25%,p16蛋白阳性表达降低,仅13.9%,强阳性表达27.8%,11例晚期卵巢癌阴性表达占30.6%;卵巢良性肿瘤p16蛋白弱阳性表达占45%,阳性表达降低占40%,对照组p16蛋白80%阳性表达,卵巢癌、卵巢良性肿瘤与正常对照比较阳性表达,P< 0.001、P< 0.05,差异显著。结论:本组卵巢上皮癌ras癌基因无突变,但ras p21过表达、p16基因25%缺失,无突变,p16蛋白表达降低,ras癌基因及p16抑癌基因异常,与卵巢癌患者临床分期、病理分化密切相关。
Objective To study the relationship between the biological behavior of ovarian carcinoma and the ras oncogene as well as p16 antioncogene. Methods The ras and p16 gene as well as their expressions were investigated in 36 specimens of ovarian carcinoma 20 benign ovarian tumor and 10 normal ovarian tissues by S-P immunohistochemistry and polymerase chain reaction single-stranded conformation polymorphism PCR-SSCP. Results No mutastion was detected in both the ovarian carcinoma specimens and the control group. Expression of ras p21 was positive in 80.6% of ovarian carcinoma weakly positive in 25% of benign ovarian tumor specimens and negative in normal ovaries P<0.01 and P< 0.001 with extremely significant differences. No mutation of p16 gene was found in ovarian carcinoma specimens and deletion of p16 was detected in 9 of 36 specimens of ovarian carcinoma and expression tests showed a decline of p16 expression in ovarian carcinoma specimens 13.9% positve 27.7% strongly positive. p16 expression was tested negative in 11 specimens 30.6% of ovarian carcinoma weakly positive in 45% and positive in 40% of benign ovarian tumors and it was positive in 80% of the control group. In contrast with the control group expression of p16 was significantly lower in the specimens of ovarian carcinoma or benign ovarian tumors P< 0.05P< 0.001. Conclusion In specimens of ovarian carcinoma from Yunnan province there was no ras genetic mutation but overexpression of ras p21 detected no other form of mutation but deletion of p16 gene was detected in 25% of specimens and its protein expression level was much lower than normal. The abnormal expressions of ras oncogene and p16 anti-oncogene were significantly associated with the clinical stage and the pathological grade of ovarian carcinoma.
