摘要
目的 研究酒精对肝脏再生的影响以及丹参对慢性酒精刺激大鼠肝脏再生的效果。方法 实验分 3组 :正常大鼠手术对照组 ;酒精组 ;丹参组。各组大鼠至 6周末行部分肝叶切除术 ,术后 2 4h全部处死 ,部分肝组织行流式细胞术 ,计算出增殖周期各期细胞百分率及增殖指数 ,部分肝组织行光镜检查。结果 酒精组与对照组及丹参组比较 ,肝脏G0 /G1期细胞百分率明显增高 ,S期和G2 M期细胞百分率明显降低 ,PI值明显降低 (P <0 .0 1和P <0 .0 5) ,肝细胞肿胀及脂滴堆积亦明显加重 ,几乎不见核分裂相 ;丹参组与对照组相比 ,G0 /G1期、S期和G2 M期细胞百分率、PI值差异均无显著性 (P >0 .0 5) 。结论 酒精能明显抑制肝脏DNA合成和再生能力 ,丹参具有促进肝脏再生的作用 。
Objective To investigate effects of chronic ethanol feeding on hepatic regeneration and the protection of Radix Salvia Mitiorrhiae (RSM) on these effects of ethanol.Methods 26 rats were divided into three groups:pair-fed group;ethanol-fed group,RSM-used group.6 weeks after the ethanol and RSM administration,All rats were performed a 70% hepatectomy and killed 24 hour later.Liver specimens were rapidly harvested for determination of liver regeneration and histological changes by Flow Cytometery (FCM) and light microscopy respectively.Results Ethanol group compared with pair and RSM group,Percentage of G 0/G 1 phase was significantly higher and Percentages of S and G 2M phases were significantly lower,PI was also significantly lower (P<0.01 and P<0.05 ,respectively);hepatocellular swelling and lipid droplet accumulation were more obvious,the karyokinesis of hepatocytes was hardly seen in ethanol group.There was no significant difference of the percentages of G 0/G 1、S、G 2M pases and PI between the pair and RSM group (P>0.05) .Conclusion The results suggest that chronic ethanol feeding can inhibit hepatocellular DAN synthesis and proliferative capacity.RSM can prevent heatocytes from alcoholic liver injury,increase liver regenerative capacity and reverse the inhibition of ethanol on liver regeneration.
