摘要 目的 探讨丹参对癫痫发作所造成神经元损伤的作用及其机制。方法 采用戊四氮致痫模型 ,运用常规病理及电镜检查方法 ,比较丹参组与对照组神经元受损的情况 ;运用免疫组织化学方法 ,对比观察丹参组与对照组热休克蛋白 (HSP) 70表达的变化。结果 丹参组与对照组动物相比 ,大脑皮质及海马区异常神经元的数目明显减少 (P<0 .0 1) ,超微结构的异常改变也明显减轻。同时丹参组动物脑内 HSP70免疫反应阳性神经元的数目明显增多 (P<0 .0 1) ,染色阳性强度亦增强。结论 丹参能减轻癫痫发作所致的神经元损伤。其作用机制可能与增加HSP70表达有关。 Objective To investigate the effect and mechanism of radix salviae miltiorrhiae(RSM) on the damages of neurous in rat brain by seizure. Methods A pentylenetetrazole (PTZ) induced seizure model was used. The extent neuronal damage and the expression of heat shock protein 70 (HSP70) were compared in the RSM group and control group by histopathological and immunohistochemistry methods. Results The number of injured neurons in the cortex and hippocampus were significantly lower in the RSM group as compared with in the control group(P<0.01). The extent of ultrastructure damage in the RSM group were milder than in the control group. The number of HSP70 immunoreactivity(IR) positive cells in the cortex and hippocampus were significantly higher in the RSM group as compared with in the control group(P<0.01),the intensity of HSP70 in the RSM group were stronger than in the control group. Conclusions RSM can protect neurons against damage by seizure. RSM probably affords neuroprotection by increase expression of HSP70.
