胃癌HER2蛋白表达及基因拷贝分析(英文)

Analysis of the Protein Expression and Gene Copy Number of HER2 in Gastric Carcinoma

摘要 目的:探讨胃癌组织HER2蛋白表达和基因拷贝数增加频率,以及蛋白表达与基因拷贝数之间的相关性。方法:分别采用免疫组织化学(IHC)和显色原位杂交(CISH)方法,检测80例青岛人胃癌组织中HER2蛋白表达和基因扩增情况。结果:HER2免疫组织化学0者51例、1+者12例、2+者12例、3+者5例。通过CISH分析:这组患者中HER2基因拷贝数扩增者共7例(8.8%),其中包括基因临界扩增3例(3.8%)。HER2的蛋白表达与基因拷贝数增加相关(P<0.05),这两个指标与肿瘤分化程度相关。HER2免疫组织化学3+和基因扩增结果一致,与乳腺癌相似。结论:这组患者中,具有较高的HER2蛋白表达和基因拷贝数增加的比率,基因扩增可能是其蛋白过表达的主要分子机制。 Objective: To investigate the frequency of the protein expression and gene copy number of HER2,and the relationship between the two markers in gastric carcinomas in Qingdao. Methods: IHC (immunohistochemistry) and CISH (Chromogenic In Situ Hy- bridization) method was used, and 80 cases protein expression and gene amplification of HER2 in gastric cancer were detected. Results: Among the 80 cases, HER2 expression was 0 in 51 tumors, I+ in 12 tumor, 2+ in 12 tumor and 3+ in 5 tumor. For CISH, seven of the 80 patients (8.8%) had increased HER2 gene copy number, including 3 patients with Critical amplification (3.8%). HER2 protein expression and genetic increased results was concordance (P〈0.05). There was a highly significant concordance between the gene increased results and IHC 3+ for HER2, which was similar to that of breast cancer. Conclusion: During this group, the patients had the higHer rate be- tween the protein expression and increased gene copy number; Gene amplification may be the major mechanism of HER-2 protein over- expression.
