摘要 目的 :阐明佐剂性关节炎 (AA)大鼠模型免疫病理特征及发病机理。方法 :应用弗氏完全佐剂诱发大鼠产生踝关节佐剂性关节炎 ,研究其免疫功能变化。结果 :AA大鼠致敏侧及对侧后肢踝关节均发生明显炎性肿胀 ,其脾细胞对SRBC诱导的抗体生成反应明显增强 ,但其脾细胞对PWM诱导的IgG的产生及血清类风湿因子 (RF)水平与对照组相比并无升高。AA大鼠对ConA诱导的脾细胞增殖反应较对照组显著降低 ,脾细胞抑制活性降低 ,CD8+ 比例较对照组增高 ,模型大鼠脾细胞IL 2mRNA表达水平较对照组明显增高 ,IFN γ ,IL 10及IL 4mRNA表达水平则明显降低。结论 :细胞免疫功能紊乱 ,尤其是Th1和Th2、Th和Ts之间的平衡紊乱是AA大鼠免疫功能异常的重要特征之一。 Objective:To study the immune characteristic and pathological feature of adjuvant arthritis rats Methods:Rat arthritis was induced by intracutaneously injection of complete Freud's adjuvant (CFA) in the right hind foot pat of rat and immune function of Adjuvant Arthritis (AA) rats was studied Results:It was found that both of the hind ankle joints were swollen after CFA injection and the antibody production response to the challenge of sheep red blood cells (SRBC) in plaque-forming cell (PFC) was significantly enhanced in AA rats However, IgG production of splenocytes induced by pokeweed (PWM) and the serum level of rheumatoid factor (RF) were not altered compared with control rats The splenocytes proliferation induced by ConA and the activity of Ts were significantly reduced, but the proportion of CD8 + of the splenocytes was markedly increased in AA rats In reverse transcription-polymerase chain reaction (RT-PCR), it was shown that the expression level of IL-2 mRNA was significantly elevated, but expression levels of IFN-γ, IL-10 and IL-4 were significantly decreased in AA rats Conclusion: Imbalance of cell-mediated immunity, especially the imbalances of the Th1/Th2 and Th/Ts, is one of the main pathological characteristics of AA rats
